Stroke remains a leading cause of death and disability worldwide. Brain edema and thromboembolic stroke are major determinants of brain damage, which can lead to the patient's death. Extravasation of plasma albumin and degradation of lipids by macrophages contribute to the formation of lipoprotein-like nanoparticles (LNPs) in the peri-infarct area. In the present study, we investigated whether LNP treatment improves brain damage after ischemic stroke and investigated the underlying mechanisms. Ischemic stroke mice were prepared by endovascular occlusion of the right middle cerebral artery and were subjected to LNP treatment. Our data showed that LNP treatment significantly reduced the infarct volume, brain edema, and neurological deficits in mice after acute ischemic stroke. It also markedly decreased the levels of interleukin (IL)-1β, IL-6, and tumor necrosis factor-α and increased the levels of IL-10 and transforming growth factor-β1. In addition, LNP treatment significantly inhibited the phosphorylation of AKT and p65 and nuclear translocation of p65 in the peri-infarct area of mice. All these changes were abolished by the co-administration of IL-10. Our findings provide the first evidence that LNP treatment exerts a protective effect against ischemic stroke injury via the IL-10-mediated inhibition of the PI3K/AKT pathway and p65 nuclear translocation.[Dysgraphia].
